Introduction

Immunocentric innovations in cancer require an integration of a Hallmark networking in order for effective treatment strategies to be developed. There are tools available that can assist with better typing, profiling and matching of the patient to determine the real-time state of the existing cancer and the requirements for developing a precision-based treatment. The single drug application model to treat a multi-factorial disease, like cancer, has fast become short-lived. Looking for a single application agent that will be the miracle drug when looking at the figure above will pretty much tell you that these days are over.

It’s time to look to the patient. The patient is the generator of the cure. The cure signature of the patient encompasses every process seen on the Hallmarks of Cancer figure above. Immunocentrics is, in fact, a patient-centric model. This model requires the ability to interrogate all data available in the data mines possible to get a more precision-based application. My theory is traditional therpaies have a high failure rate due to the inferior typing, profiling and matching of the patient. The patient is the generator.

The Hallmarks

There are ten Hallmarks of cancer as seen in the above figure. The expanded controls of each of the ten categories is crucial in understanding how a cancer is behaving from the core DNA expression, integral genetic / epigenetics, cell cycling and programming, metabolism, inflammation signaling, activation and induction, immune defense and regulation, evasion, resistance and immortalization. Mutation, migration and proliferation are all influenced by a combination of these Hallmarks and can be cohesive to one another for the good or the bad responses seen in cancer progression or regression.

Analytics / Bioinformatics A Necessary Patient Baseline

Cancer Hallmarks Analytics Tool (CHAT) is a comprehensive text mining approach that interrogates research literature for every cancer type. In patient profiling, it is essential in laying the framework for treatment development specific to the patient. An example of this is developing molecular-matched precision products specific to the patient’s Hallmark expressions. https://academic.oup.com/bioinformatics/article/33/24/3973/3965324

Immunocentrics

Immunocentrics is a system that organizes the specialties of immunogenetics, epigenetics, proteomics, nanoengineering, bioinformatics, biostatistics, immuno-sequencing and synthesis of peptides and antibodies and development of precision-based personalized cancer vaccines from concept innovation to bedside. The integration, coordination and cooperation of these specialties is fundamental in delivering precision-based patient specific immunotherapy in cancer treatment. Immunocentrics integrates the Hallmark profiling in a patient centered application. https://www.technion.ac.il/en/2018/06/the-immune-system-cracking-open-the-black-box/

Summary

In the webinar, “Methylation and Cancer,” I briefly discuss the basics of why an integration model such as this is necessary. Over a 20 year period, I have treated numerous patients with aggressive end-stage cancers implementing this integration model with sucess and a high curative response. https://www.youtube.com/watch?v=xy5S1cEfS9A
